Anticancer effects of isothiocyanates and their combination with other therapeutic approaches.

Proposed project is aimed at basic research of anticancer effects of isothiocyanates (ITCs). We will pursue primarily in vitro studies on human hematopietic and carcinoma cell lines with the goal of identifying endpoints for subsequent in vivo experiments. Based on our and others’ previous studies, the investigation of ITC synergism in new treatment combinations will be continued. With the aid of flow cytometry, real-time PCR, biochemical and immunochemical methods we will follow principal events, signaling pathways and factors influencing synergy of ITCs with cisplatin- and anthracycline- based chemotherapeutics. Parallelly we will test the new possibilities of ITC combinations with novel experimental therapeutic approaches such as suicide gene therapy and nanosized particle drugs. In the second part we aspire to prove the anticancer effects of SFN in vivo, following its efficacy on tumor growth, formation of experimental metastasis, cytokine production, chromosomal aberations, DNA repair and gene expression
